I hate shit like this. Too many.

Take SouthPark. HAS to be Cartman. But goddamnit if Randy isn't one of the funniest characters ever.

Archer...another good example, where the titular character gets the nod, but Krieger and Tunt are the funniest characters IMO.

The vast majority of my choices would be cartoons. I'll give nod to real characters, but they will likely be old.
