So:

1) Clemson is the #1 overall team in the nation
2) Clemson is playing at a favorable location
3) Clemson beat Oklahoma 40-6 last time they played with about 75% of the same players
4) Oklahoma hasnt beaten a team with their starting quarterback in a month
5) Clemson has the best QB, OU has faced all year by a large margin, and the only starting QB as well

Why wouldn't Clemson be confident?
Don't see why not as long as they think this is the same OU team they beat last year, but things have changed. For one, Brent V. was very familiar with J. Heuple and his play calling (may have even known some of the signals) and what he would try to do. Second, Knight was QB last time with an injured Sheppard trying to play. Fact is, B.V. was never really very great at defending a spread attack (one reason why he is no longer at OU), last year wasn't really a spread by OU, more of a zone read. Now, Riley has a real offense to work with and a QB that knows how to operate it, plus, Sheppard isn't the only WR on the field that can play and two RB's that have to be accounted for. With an offense that can give the defense some rest this time around don't expect the same outcome as last year.

Clemson hasn't faced an offense or a QB like Mayfield yet either, should be interesting.

Kizer and Marquise Williams are nothing to squeeze at.
Kizer is 15th highest QBR averaging 80.0 but against Clemson: 58.4
Williams is 21st highest QBR averaging 77.1 but against Clemson: 24.4

Sorry Oklahoma, Clemson is not another BIG 12 defense.

I mean there are 6 teams ahead of Oklahoma in total defense.

Imo total defense isn't necessarily the best way to evaluate defense as it gives too much loves to teams that play in conferences that don't have high octane offenses. I'll have to look at some numbers later when I have time to really make an informed post on this.

Well, North Alabama were heavy favorites over Tuskegee a couple of weeks back in the D2 playoffs, and the Lions fell.

Hopefully, that's no indicator for a favored Tide over MSU.
